凈(jing)化(hua)工(gong)作(zuo)(zuo)(zuo)臺(tai)(tai)(tai)是一種局部層流(liu)裝置,能在局部形成高(gao)潔度的(de)(de)(de)工(gong)作(zuo)(zuo)(zuo)環(huan)境。它由(you)工(gong)作(zuo)(zuo)(zuo)臺(tai)(tai)(tai)、過濾(lv)器(qi)、風機、靜壓箱和支撐(cheng)體等(deng)組(zu)成,采用過濾(lv)空氣(qi)(qi)使工(gong)作(zuo)(zuo)(zuo)臺(tai)(tai)(tai)操(cao)作(zuo)(zuo)(zuo)區達到凈(jing)化(hua)除(chu)(chu)菌的(de)(de)(de)目的(de)(de)(de)。室內空氣(qi)(qi)經(jing)預過濾(lv)器(qi)和高(gao)效(xiao)過濾(lv)除(chu)(chu)塵后以(yi)垂直(zhi)或水(shui)平層流(liu)狀態通過工(gong)作(zuo)(zuo)(zuo)臺(tai)(tai)(tai)的(de)(de)(de)操(cao)作(zuo)(zuo)(zuo)區,由(you)于(yu)空氣(qi)(qi)沒有渦流(liu),所以(yi),任何一點灰塵或附(fu)著在灰塵上的(de)(de)(de)雜菌都能被(bei)排除(chu)(chu),不(bu)易(yi)向別處(chu)擴散和轉移。因此,可使操(cao)作(zuo)(zuo)(zuo)區保持無菌狀態。
與無茵檢測室和疫苗接種箱對比,施用靜化業務臺存在業務必備條件好、基本操作便捷、無茵檢測功能可靠的、無消毒殺菌解毒劑對人體肌肉害處、占建筑面積小且可可移動等好處。若果擺放在無茵檢測空間內施用,無茵檢測功能更強。其優點缺點是價格多少最貴,預篩選器和提高效率篩選器還須要定時清潔工作和更換。 高壓力空氣壓縮殺菌鍋高(gao)壓蒸(zheng)(zheng)汽(qi)滅(mie)菌鍋(guo)是一(yi)個密閉的、可以(yi)耐受一(yi)定壓力的雙層(ceng)金(jin)屬鍋(guo)。鍋(guo)底或夾層(ceng)內(nei)盛水,當(dang)水在(zai)(zai)鍋(guo)內(nei)沸(fei)騰時由(you)于蒸(zheng)(zheng)汽(qi)不(bu)能逸出,使鍋(guo)內(nei)壓力逐漸升高(gao),水的沸(fei)點和(he)溫度(du)可隨之升高(gao),從(cong)而達(da)到高(gao)溫滅(mie)菌的目的。一(yi)般(ban)在(zai)(zai)0.11MPa的壓力下,121℃滅(mie)菌20~30min,包括芽孢在(zai)(zai)內(nei)的所有(you)微(wei)生(sheng)物均可被(bei)殺死。如果(guo)滅(mie)菌物品體積較大(da),蒸(zheng)(zheng)汽(qi)穿透(tou)困(kun)難,可以(yi)適(shi)當(dang)提(ti)高(gao)蒸(zheng)(zheng)汽(qi)壓力或延長滅(mie)菌時間(jian)。
油田過濾除菌鍋有臥式、直列V型、手攜式式等多種多樣類型的,在微怪物學試驗室,極為較為常用的是手攜式式和直列V型油田蒸汽發生器過濾除菌鍋。和自然壓過濾除菌鍋想必,油田過濾除菌鍋的獨到之處是過濾除菌必需的準確時間太快、浪費助燃劑、過濾除菌徹底刪除文件等。其優點缺點是價格非常昂貴,過濾除菌余量較小。 鍛煉箱培養箱(xiang)(xiang)(xiang)(xiang)是培養微(wei)(wei)生物(wu)的(de)(de)專用(yong)(yong)設備。制(zhi)熱式培養箱(xiang)(xiang)(xiang)(xiang)是由(you)電爐絲和(he)溫(wen)度(du)控制(zhi)儀合成的(de)(de)固定體(ti)積的(de)(de)恒(heng)溫(wen)培養裝置(zhi),大小規格不一(yi)。微(wei)(wei)生物(wu)實驗(yan)室常用(yong)(yong)的(de)(de)培養箱(xiang)(xiang)(xiang)(xiang)工作容積有450×450×350mm3或650×500×500mm3,適用(yong)(yong)于室溫(wen)至60℃之間的(de)(de)各(ge)類微(wei)(wei)生物(wu)培養。目前(qian),隨著科學水平的(de)(de)發展,培養箱(xiang)(xiang)(xiang)(xiang)設備的(de)(de)完善程(cheng)度(du)和(he)價格有很大差(cha)別(bie)。有各(ge)種結構合理(li)、功(gong)能齊全的(de)(de)培養箱(xiang)(xiang)(xiang)(xiang),如恒(heng)溫(wen)培養箱(xiang)(xiang)(xiang)(xiang)、恒(heng)溫(wen)恒(heng)濕培養箱(xiang)(xiang)(xiang)(xiang)、低溫(wen)培養箱(xiang)(xiang)(xiang)(xiang)、微(wei)(wei)生物(wu)多用(yong)(yong)培養箱(xiang)(xiang)(xiang)(xiang)和(he)二氧化碳(tan)培養箱(xiang)(xiang)(xiang)(xiang)等。有的(de)(de)用(yong)(yong)計算機控制(zhi),可(ke)選擇多條時間線變(bian)換(huan)溫(wen)差(cha),從而克服了環境(jing)溫(wen)度(du)的(de)(de)影響(xiang),一(yi)年(nian)四(si)季均能達到培養要求(qiu)的(de)(de)溫(wen)度(du)。
微生(sheng)物(wu)多用培養(yang)箱是(shi)集加熱(re)、制冷和(he)振蕩(dang)于一(yi)體的微生(sheng)物(wu)液體發酵裝(zhuang)置。工作室的溫(wen)(wen)度在(zai)15~50℃范(fan)圍內(nei)(nei)任(ren)意選(xuan)定,選(xuan)定后經(jing)溫(wen)(wen)控(kong)(kong)儀自動(dong)控(kong)(kong)制,保持工作室內(nei)(nei)恒溫(wen)(wen)。同(tong)時設有可控(kong)(kong)硅調(diao)速系統,振蕩(dang)機轉速可在(zai)1~220rpm范(fan)圍內(nei)(nei)任(ren)意調(diao)控(kong)(kong)。
潮濕箱干燥(zao)(zao)箱(xiang)(xiang)(xiang)(xiang)是用于除(chu)去潮(chao)濕物料內(nei)及器皿內(nei)外水分或其它揮發性溶液的(de)(de)設備。類型(xing)(xing)很多(duo),有(you)(you)(you)箱(xiang)(xiang)(xiang)(xiang)式、滾筒式、套間(jian)式、回轉式等(deng)。微生物學實(shi)驗室(shi)(shi)多(duo)用箱(xiang)(xiang)(xiang)(xiang)式干燥(zao)(zao)箱(xiang)(xiang)(xiang)(xiang),大小規格不(bu)一。工作室(shi)(shi)內(nei)配有(you)(you)(you)可活動的(de)(de)鐵(tie)絲網板,便(bian)于放置被干燥(zao)(zao)的(de)(de)物品(pin)。制熱(re)(re)升溫(wen)(wen)式干燥(zao)(zao)箱(xiang)(xiang)(xiang)(xiang)也是有(you)(you)(you)電爐絲和溫(wen)(wen)度(du)控(kong)制儀(yi)組(zu)(zu)成,可調節溫(wen)(wen)度(du)從室(shi)(shi)溫(wen)(wen)至(zhi)300℃任意選擇。有(you)(you)(you)的(de)(de)干燥(zao)(zao)箱(xiang)(xiang)(xiang)(xiang)采用導電溫(wen)(wen)度(du)計為(wei)敏感元(yuan)件,配合晶體管和繼電器組(zu)(zu)成自動控(kong)制系統,克服了金屬管型(xing)(xing)熱(re)(re)膨脹控(kong)制的(de)(de)缺點。此外,還有(you)(you)(you)真空(kong)干燥(zao)(zao)箱(xiang)(xiang)(xiang)(xiang)(配有(you)(you)(you)真空(kong)泵和氣壓表),可在常壓或減壓下操作。
搖床
搖(yao)(yao)床(chuang)(chuang)(chuang)又稱搖(yao)(yao)瓶(ping)機,它是培養好氣(qi)性微(wei)生(sheng)物的(de)(de)小型試(shi)驗設備或(huo)作為(wei)種子擴大培養之(zhi)用,常用的(de)(de)搖(yao)(yao)床(chuang)(chuang)(chuang)有往復式和旋轉式兩種。往復式搖(yao)(yao)床(chuang)(chuang)(chuang)的(de)(de)往復頻率(lv)一(yi)(yi)般在80~140次(ci)/min,沖(chong)程(cheng)一(yi)(yi)般為(wei)5~14cm,如(ru)頻率(lv)過(guo)快、沖(chong)程(cheng)過(guo)大或(huo)瓶(ping)內(nei)液(ye)體(ti)裝量過(guo)多,在搖(yao)(yao)動(dong)時液(ye)體(ti)會(hui)濺到包扎瓶(ping)口的(de)(de)紗布或(huo)棉塞上,導致雜菌(jun)污染,特別(bie)是啟動(dong)時更容易(yi)發生(sheng)這種情況。旋轉式搖(yao)(yao)床(chuang)(chuang)(chuang)的(de)(de)偏心(xin)距(ju)一(yi)(yi)般在3~6cm之(zhi)間,旋轉次(ci)數為(wei)60~300rpm。
放在搖床上的(de)(de)(de)培(pei)養瓶(ping)(ping)(一(yi)般為三(san)(san)角瓶(ping)(ping))中的(de)(de)(de)發酵(jiao)液所需要(yao)的(de)(de)(de)氧是由空(kong)氣經瓶(ping)(ping)口(kou)包扎的(de)(de)(de)紗布(bu)(一(yi)般8層)或棉塞(sai)通(tong)入的(de)(de)(de),所以氧的(de)(de)(de)傳遞與(yu)瓶(ping)(ping)口(kou)的(de)(de)(de)大小、瓶(ping)(ping)口(kou)的(de)(de)(de)幾(ji)何形狀、棉塞(sai)或紗布(bu)的(de)(de)(de)厚(hou)度和密度有關。在通(tong)常情況下,搖瓶(ping)(ping)的(de)(de)(de)氧吸收系數取決于搖床的(de)(de)(de)特性和三(san)(san)角瓶(ping)(ping)的(de)(de)(de)裝(zhuang)樣量。

疫苗注射箱接種箱(xiang)分為(wei)固體菌種接種箱(xiang)和液體菌種接種箱(xiang)兩(liang)(liang)種。固體菌種接種箱(xiang)是一(yi)(yi)個用木料(liao)和玻璃制成或(huo)(huo)由(you)有機玻璃焊接而成的(de)密閉(bi)小(xiao)箱(xiang)。又分為(wei)雙人和單人操(cao)作(zuo)箱(xiang)。箱(xiang)體可大可小(xiao),一(yi)(yi)般(ban)箱(xiang)體長約143cm,寬86cm,總(zong)高154cm,支(zhi)架76cm。箱(xiang)的(de)上(shang)部左右兩(liang)(liang)側(ce)各裝有兩(liang)(liang)扇能啟閉(bi)的(de)玻璃推拉(la)門,方便菌種進(jin)(jin)出。窗(chuang)的(de)下部分別設有兩(liang)(liang)個直徑(jing)約13cm的(de)圓(yuan)洞,兩(liang)(liang)洞的(de)中(zhong)心距離為(wei)52cm(同肩寬),洞口(kou)裝有帶(dai)(dai)松(song)緊帶(dai)(dai)的(de)袖(xiu)套,以防(fang)雙手(shou)在箱(xiang)內(nei)(nei)操(cao)作(zuo)時,外界空氣進(jin)(jin)入箱(xiang)內(nei)(nei)造成污染。操(cao)作(zuo)時兩(liang)(liang)人相對而坐,雙手(shou)通過(guo)袖(xiu)套伸入箱(xiang)內(nei)(nei)。箱(xiang)兩(liang)(liang)側(ce)最(zui)好也裝上(shang)玻璃,箱(xiang)頂部為(wei)木板或(huo)(huo)玻璃。箱(xiang)內(nei)(nei)頂部裝有紫外線殺(sha)菌燈和照(zhao)明用日(ri)光燈各一(yi)(yi)支(zhi)。箱(xiang)體安裝木板或(huo)(huo)玻璃均可,但要注意密封。
液體菌種(zhong)(zhong)接種(zhong)(zhong)箱(xiang)(xiang)是專(zhuan)為(wei)移(yi)接液體菌種(zhong)(zhong)而設計的。比(bi)固體菌種(zhong)(zhong)箱(xiang)(xiang)窄長(chang),單側兩人(ren)操(cao)作。內設軌道和紫(zi)外線燈,箱(xiang)(xiang)兩端開有(you)高25cm,寬10cm的長(chang)方形出(chu)口,方便菌種(zhong)(zhong)進出(chu),洞口設有(you)小(xiao)(xiao)推門(men)。進出(chu)口下處設蒸汽源,接種(zhong)(zhong)時用蒸汽封(feng)住進出(chu)口,以防雜菌進入箱(xiang)(xiang)內。箱(xiang)(xiang)背面設有(you)液體菌種(zhong)(zhong)移(yi)接管能進入的小(xiao)(xiao)孔。
接種(zhong)箱滅(mie)(mie)菌(jun)時(shi)(shi)(shi),用紫外(wai)線(xian)照射30min。如(ru)果沒有紫外(wai)線(xian)燈,可用甲醛和(he)(he)高(gao)錳酸鉀(甲醛10~14mL/m3+高(gao)錳酸鉀5~7g/m3空間)熏(xun)蒸30min以上。使用時(shi)(shi)(shi),先將(jiang)所需(xu)物品(pin)和(he)(he)工具(ju)放入接種(zhong)箱內,然后進(jin)(jin)行藥(yao)劑熏(xun)蒸和(he)(he)紫外(wai)線(xian)滅(mie)(mie)菌(jun),再按(an)無(wu)菌(jun)操作進(jin)(jin)行接種(zhong)。接種(zhong)箱的結構(gou)簡單,造(zao)價(jia)低(di)廉,易消毒滅(mie)(mie)菌(jun),操作方便,而且人(ren)在箱外(wai)操作,氣溫(wen)較高(gao)時(shi)(shi)(shi)也能(neng)作業。缺點是(shi)進(jin)(jin)出培養基費工費時(shi)(shi)(shi),每次接種(zhong)前(qian)都需(xu)要進(jin)(jin)行滅(mie)(mie)菌(jun)。
電冰箱 微生物實驗室的冰箱主要有兩種:普通冰箱和低溫冷凍冰箱。普通冰箱一般都具有兩個柜子,即鮮藏柜和冷藏柜,溫度分別為4℃和-20℃;低溫冷凍冰箱溫度一般控制在-40~-80℃。它們都可以用于微生物菌種保藏。鮮藏柜常用于保存斜面菌種,保藏時間在3個月左右。超過3個月,斜面就會變干,因此需要轉接菌種。如果要長時間保存菌種,則需要經過處理后,貯藏于普通冰箱的冷藏柜或低溫冷凍冰箱中,它們的保藏時間較長,一般都在1年以上。
